You can, just not via the web site. I do it in Tweetdeck all the time.

You can also do it via the website, sort of.

If a tweet is a reply to another one, there is a "in reply to" link right under the tweet. I guess other client applications (like tweetdeck) use this link to create a conversation view.

I suppose, it is quite similar to what happens with e-mail.
