In this case I suspect you've set up the buildbot on a completely new Windows install. In which case you're probably missing the proper Microsoft C Runtime. (Windows update would eventually get to this). According to the docs : https://docs.python.org/3.4/using/windows.html Python 3.5 has a dependency on the VC2015 runtime https://www.microsoft.com/en-us/download/details.aspx?id=48145 and Python 3.4 on the VC2008 runtime https://www.microsoft.com/en- us/download/details.aspx?id=15336 The load is probably failing because ctypes loads a dll that needs the runtime.
